Description
Here's the condensed version:
Tesamorelin
Tesamorelin is a synthetic 44-amino acid polypeptide analog of growth hormone-releasing hormone (GHRH), with modifications at both the N- and C-terminus to enhance stability and resistance to enzymatic degradation. By interacting with GHRH receptors in the anterior pituitary gland, it may stimulate growth hormone secretion and downstream IGF-1 production via the cAMP-PKA signaling pathway. Research suggests it may produce an estimated 69% increase in overall GH levels and a 122% surge in IGF-1 — without appearing to alter GH pulse frequency or peak levels.

Research Highlights
- Lipodystrophy — Two Phase III studies across 806 subjects over 26 weeks found Tesamorelin-exposed models exhibited at least a 15.4% reduction in visceral adipose tissue, alongside significant decreases in triglyceride and cholesterol levels compared to placebo.
- Hepatic Fat — A 12-month study in 61 immunodeficient subjects with elevated hepatic fat fractions found 35% of Tesamorelin subjects showed meaningful reductions in liver fat vs. only 4% in the placebo group, with no reported impact on glucose levels.
- Visceral Fat — Research in lipodystrophy models suggests Tesamorelin may reduce visceral fat accumulation by up to 25%, potentially addressing associated metabolic complications including insulin resistance, elevated LDL, and atherosclerosis risk.
- Muscle Tissue — CT scan analysis indicated potential improvements in density and volume of specific muscle groups — including the rectus abdominis, psoas major, and paraspinal muscles — alongside reductions in intramuscular fat compared to placebo.
- Insulin Sensitivity — A 12-week randomized trial in 53 Type II diabetes subjects found no significant alteration in fasting glucose, glycosylated hemoglobin, or diabetes control markers across all dose groups.
- Cognition — An ongoing clinical study in immunodeficient subjects with mild cognitive impairment is evaluating Tesamorelin's effect on neurocognitive performance via Global Deficit Score at 6 and 12 months; final results are pending.
