Description
GLP-2 is a synthetic peptide and dual hormone receptor agonist targeting the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. Built on a GIP-based sequence with a C20 fatty diacid side chain that supports albumin binding and a long half-life, it was the first dual GIP/GLP-1 agonist to be studied at scale and remains one of the most widely researched compounds in metabolic and obesity research.

Research Highlights
- Weight Reduction — Phase 3 clinical trials reported average body weight reductions of up to roughly 20% over 72 weeks at higher doses, exceeding results observed with GLP-1 single agonists in comparable populations.
- Glycemic Control — Research in type 2 diabetes models reported substantial reductions in HbA1c, with GIP and GLP-1 agonism proposed to act together on glucose-dependent insulin secretion.
- Dual Agonism — Studies suggest the compound is imbalanced in favor of the GIP receptor, and that GIP receptor engagement may contribute to effects beyond those seen with GLP-1 agonism alone.
- Lipid and Cardiometabolic Markers — Trial data suggested possible improvements in triglycerides, blood pressure, and other cardiometabolic markers alongside weight reduction.
- Hepatic Fat — Research has examined the compound in models of metabolic-dysfunction associated fatty liver disease, with reported reductions in liver fat content.
